I reckon you can get polyploidy without colchicine, with the right sort of parental chromosomal combinations (though how that's determined is something I have no idea about). I do believe it has to do with differentiation though, so you need sort of complementary but different parents...based on pure speculation.
Signs of polyploidy, afaik, is general "oversized" sort of features. I definitely have some strains that are rather freakish in that sense, and they were from almost random pollination. I say almost, because there was crude selection involved. If they're actually polyploid, I dunno.
