Do I need to defoliate again? Are these buds/leaves at rhe bottom needed? Or should I remove them?

Let her grow, if some big leaf shades cut it at half. Plant need energy for buds, from leaves when you cut off them she will developing new leaves and buds will be smaller. Just cut leaf at half if you need, bud she is OK. And second answear is you gave 3 weeks to go
