When to defoliate in Flower?

When readings say defoliate after 21 days - is that from flipping to 12/12 or until you notice the plant has gone into flower?

Hi. The flowering period only begins with the first signs of blooming. When you switch to 12/12, the plant just releases the hormones that trigger it to go into flowering. But at 21 days after 12/12, you're not doing anything wrong.
