Remember, For every molecule of glucose produced during photosynthesis, a plant needs to split six molecules of water. This process provides the hydrogen needed for synthesizing glucose and other organic compounds, while oxygen is released as a byproduct. Homework. If Rubisco activity is impaired and it cannot properly function or regenerate its substrate, the plant's leaves are likely to turn a pale green or lime green, a condition known as chlorosis. Essentially, Rubisco activity is highly regulated and susceptible to various environmental and metabolic factors that can cause it to become inhibited, leading to an apparent failure in RuBP regeneration due to a lack of consumption. Rubisco regeneration is intrinsically linked to nitrogen supply because Rubisco is a major sink for nitrogen in plants, typically accounting for 15% to over 25% of total leaf nitrogen. The regeneration phase itself consumes nitrogen through the synthesis of the Rubisco enzyme and associated proteins (like Rubisco activase), and overall nitrogen status heavily influences the efficiency of RuBP regeneration. RuBisCO is a very large enzyme that constitutes a significant proportion (up to 50%) of leaf soluble protein and requires large investments in nitrogen. Insufficient nitrogen supply limits the plant's ability to produce adequate amounts of RuBisCO, thereby limiting the overall capacity for photosynthesis and carbon fixation. Maintaining the optimal, slightly alkaline pH is crucial for the proper function and regeneration of Rubisco. Deviations in either direction (too high or too low) disrupt the enzyme's structure, activation state, and interaction with its substrates, leading to decreased activity and impaired RuBP regeneration. (Lime/yellowing) Structural Component: Nitrogen is an essential building block for all proteins, and the sheer abundance of the Rubisco protein makes it the single largest storage of nitrogen in the leaf. Synthesis and Activity: Adequate nitrogen supply is crucial for the synthesis and maintenance of sufficient Rubisco enzyme and Rubisco activase (Rca), the regulatory protein responsible for maintaining Rubisco's active state. Nitrogen deficiency leads to a decrease in the content and activity of both Rubisco and Rca, which in turn limits the maximum carboxylation rate, Vmax, and the rate of RuBP regeneration Jmax, thus reducing overall photosynthetic capacity. Nitrogen Storage and Remobilization: Rubisco can act as a temporary nitrogen storage protein, which is degraded to remobilize nitrogen to other growing parts of the plant, especially under conditions of nitrogen deficiency or senescence. Nitrogen Use Efficiency (NUE): The allocation of nitrogen to Rubisco is a key determinant of a plant's photosynthetic nitrogen use efficiency (PNUE). In high-nitrogen conditions, plants may accumulate a surplus of Rubisco, which may not be fully activated, leading to a lower PNUE. Optimizing the amount and activity of Rubisco relative to nitrogen availability is a target for improving crop NUE. Photorespiration and Nitrogen Metabolism: Nitrogen metabolism is also linked to the photorespiration pathway (which competes with carboxylation at the Rubisco active site), particularly in the reassimilation of ammonia released during the process. To increase RuBisCO regeneration, which refers to the process of forming the CO2 acceptor molecule Ribulose-1,5-bisphosphate (RuBP) during photosynthesis, the primary methods involve optimizing the levels and activity of Rubisco activase (Rca) and enhancing the performance of other Calvin-Benson-Bassham (CBB) cycle enzymes. Biochemical and Environmental Approaches: Optimize Rubisco Activase (Rca) activity: Rca is a crucial chaperone protein that removes inhibitory sugar phosphates, such as CA1P (2-carboxy-D-arabinitol 1-phosphate), from the Rubisco active site, thus maintaining its catalytic competence. •Ensure optimal light conditions: Rca is light-activated via the chloroplast's redox status. Adequate light intensity ensures Rca can effectively maintain Rubisco in its active, carbamylated state. •Maintain optimal temperature: Rca is highly temperature-sensitive and can become unstable at moderately high temperatures (e.g., above 35°C/95F° in many C3 plants), which decreases its ability to activate Rubisco. Maintaining temperatures within the optimal range for a specific plant species is important. •Optimize Mg2+ concentration: Mg2+ is a key cofactor for both Rubisco carbamylation and Rca activity. In the light, Mg2+ concentration in the chloroplast stroma increases, promoting activation. •Manage ATP/ADP ratio: Rca activity depends on ATP hydrolysis and is inhibited by ADP. Conditions that maintain a high ATP/ADP ratio in the chloroplast stroma favor Rca activity. Enhance Calvin-Benson-Bassham (CBB) cycle enzyme activity: The overall rate of RuBP regeneration can be limited by other enzymes in the cycle. •Increase SBPase activity: Sedoheptulose-1,7-bisphosphatase (SBPase) is a key regulatory enzyme in the regeneration pathway, and increasing its activity can enhance RuBP regeneration and overall photosynthesis. •Optimize other enzymes: Overexpression of other CBB cycle enzymes such as fructose-1,6-bisphosphate aldolase (FBA) and triose phosphate isomerase (TPI) can also help to balance the metabolic flux and improve RuBP regeneration capacity. Magnesium ions, Mg2+, are specifically required for Rubisco activation because the cation plays a critical structural and chemical role in forming the active site: A specific lysine residue in the active site must be carbamylated by a CO2 molecule to activate the enzyme. The resulting negatively charged carbamyl group then facilitates the binding of the positively charged Mg2+ion. While other divalent metal ions like Mn2+ can bind to Rubisco, they alter the enzyme's substrate specificity and lead to dramatically lower activity or a higher rate of the non-productive oxygenation reaction compared to Mg2+, making them biologically unfavorable in the context of efficient carbon fixation. The concentration of Mg2+ in the chloroplast stroma naturally increases in the light due to ion potential balancing during ATP synthesis, providing a physiological mechanism to ensure the enzyme is activated when photosynthesis is possible. At the center of the porphyrin ring, nestled within its nitrogen atoms, is a Magnesium ion (Mg2+). This magnesium ion is crucial for the function of chlorophyll, and without it, the pigment cannot effectively capture and transfer light energy. Mg acts as a cofactor: Mg2+ binds to Rubisco after an activator CO2 molecule, forming a catalytically competent complex (Enzyme-CO2-Mg2+). High light + CO2) increases demand: Under high light (60 DLI is a very high intensity, potentially saturating) and high CO2, the plant's capacity for photosynthesis is high, and thus the demand for activated Rubisco and the necessary Mg2+ cofactor increases. Mg deficiency becomes limiting: If Mg2+ is deficient under these conditions, the higher levels of Rubisco and Rubisco activase produced cannot be fully activated, leading to lower photosynthetic rates and potential photo-oxidative damage. Optimal range: Studies show that adequate Mg2+ application can enhance Rubisco activation and stabilize net photosynthetic rates under stress conditions, but the required concentration is specific to the experimental setup. Monitoring is key: The most effective approach in a controlled environment is to monitor the plant's physiological responses e.g., leaf Mg2+ concentration, photosynthetic rate, Rubisco activation state, and adjust the nutrient solution/fertilizer to maintain adequate levels, rather than supplementing a fixed "extra" amount. In practice, this means ensuring that Mg2+ is not a limiting factor in the plant's standard nutrient solution when pushing the limits with high light and CO2. Applying Mg2+ through foliar spray is beneficial to Rubisco regeneration, particularly in alleviating the negative effects of magnesium (Mg) deficiency and high-temperature stress (HTS). While Mg can be leached from soil, within the plant it is considered a mobile nutrient, particularly in the phloem. Foliar-applied Mg is quickly absorbed by the leaves and can be translocate to other plant parts, including new growth and sink organs. Foliar application of: NATURES VERY OWN MgSO4 @ 15.0g L-1 in a spray bottle. For those high-intensity workouts when 1 meal a day is just not enough! Foliar sprays are often recommended as a rapid rescue measure for existing deficiencies or as a supplement during critical growth stages, when demand for Mg is high. Application in the early morning or late evening can improve absorption and prevent leaf burn. The plant was getting a little limey yellow in the centre. Shortly thereafter, she was back in business, green mostly regenerated. The starting point [of creativity] is curiosity: pondering why the default exists in the first place. We’re driven to question defaults when we experience vuja de, the opposite of déjà vu. Déjà vu occurs when we encounter something new, but it feels as if we’ve seen it before. Vuja de is the reverse—we face something familiar, but we see it with a fresh perspective that enables us to gain new insights into old problems. Confidence is evidence... nothing more. You are confident because you have driven 10,000 times, you are confident because you have spoken 10,000 times. People think confidence is a feeling, but it's not. If you want more confidence, then you need to create evidence, take more shots, collect more data, build more experiences, take more risks; fail, confidence doesn't come first; it is the reward you get for doing the work. no one else wants to do.

11-29 Bad news, Banana Purple Punch has been let go. 😭 She was very stunted and tiny, so I made the bold decision to call it. Thought about letting her finish, but she was really small. But Strawberry cheesecake is making up for it!. This thing seems super happy atm! She has jumped to around 12 inches. Stretch is officially on! Triple grape is rockin too, few flimsy leafs, bud branches. One of the branches, kinda twisted itself. Imma look into that one. But both rockin and lovin the EM1. More importantly than the rest! I still haven't over watered or fed. 😁💪 Time will tell, but I'm staying the course! Happy Holidays 😁🤘 12-1 2k water, silica 3ml, cal/mag 3ml, drops balance 1ml and some dust of yucca. 6.2 Stripped GC of anything blocking anything else . Couple blocked bud sites too. Wont touch for at least 3 days. Maybe 4, we will say next water? She may be a bit stressed 😂 Im sure the stripping didnt help. 💪🤘 3x- 11.5 inches tall Scc- 12.5 in tall . .

Day 86 12/05/2021- Week 5 of flower almost done hopefully by the end of the month. Went out on a trip and GoPro never turn on for the last two days bummer but will upload time lapse videos daily. Today the lady got nutrients as well. Day 89 12/08/2021- Today the lady got water, really hoping to see buds starting to fatten up. Day 91 12/10/2021- So today the lady got a special treat, freshly collected rain water. She is just doing her thing and making those buds. I have also finally put the spider farmer SF-2000 at 100%. 4/22/2024 Day 29 showing plenty pistils, still in the transition to flower stage. Training may help the plant grow bigger/ longer stems. They can be semi bushy when spread out. Switched nutes to flower bloom from Gaia green 4/26 day 33? Last half of week 5 there has been more of a stretch than there was previous to this. These plants have been mad with a bit of over water and chlorine issues and maybe cal mag 4/27 day 34 end of week 5 The plants both stretched. The one I didn’t train,I can see a visible growth of about double the height, just over this last week. For it being just a pre flower stretch/transition phase for the auto flowers, I am surprised at how many pistils are on the flower sights. The end of last week, I didn’t feel like there were enough pistils to consider it the beginning of flower, and now at the end of this week, it looks like this should be the end of week 1 flower, but they were still stretching.

**Week 8 - Third Week of Flower** Things are getting wild in the Donutz tent! After the insane stretch last week, the plants have turned the space into a full-on jungle. The start of Week 8 called for drastic measures, and the results are setting the stage for explosive bud production. **Key Updates**: - **Aggressive Defoliation**: On Day 1 of the third flower week, the plants underwent a *brutal* defoliation. Every bud site that had started to develop its own leaves was stripped of its fan leaves, allowing light to flood the canopy. Even the smallest sites now have excellent exposure, giving them the best chance to mature into chunky nugs in the coming weeks. Despite the intensity of the trim, the plants are resilient and primed to bounce back strong. - **Boosted Nutrients**: To support the plants after the stress of defoliation, I added another 5-6g of Greenhouse Feeding Bloom per plant. This ensures the soil stays nutrient-rich and gives the plants the resources they need to recover and thrive. - **Watering**: Still rocking reverse osmosis (RO) water, ensuring clean and efficient hydration without any surprises. **Observations**: - The canopy is significantly cleaner and more open, with light now reaching deeper into the lower sections. - The plants are handling the defoliation well, with no immediate signs of stress or shock. - Bud sites are already looking promising, with visible development and an even distribution across the canopy. **Looking Ahead**: The next few weeks will be all about letting the plants recover and watching the buds stack up. With improved light penetration and a fresh dose of nutrients, everything is lined up for some impressive development. The Donutz are thriving, and the Florida Gang vibes are at an all-time high — the tent is set to deliver big!

In general: Red Pure Auto CBD 💚❤️💚❤️ - It is a beautiful plant. It still has deep green leaves, those that are close to the flowers are dark purple. It is clear that the flowers will be dark red, even purple. The plant responds well to everything from temperature, humidity, to excellent nutrient response, especially during the transition to flowering. It also responded well to the nutrient change in general. The truth is that I was very careful. Anyway, it looks really healthy and is enjoying life💚🌺🌱 Training: I did LST only on two small branches, which were blocked from the light by older strong branches and also a slight defoliation. It was really a few leaves that cast a shadow on the buds 💚🌞🌿🌞 Light: Mars Hydro TS1000 The plants respond effectively and the light is technically very well managed - I try to have 50 cm between the light and the plants, so I move it up regularly 💡💡💡 Thanks for the likes See you soon 😍😍

I didn't feed again this week due to it being the same nutrients. My ec was running 3.9 so I added just regular ro water to get it closer to 2.4ec.

Day 120- Day 72- 11/11 - Still just giving that liter of plain, non pH'd RO water every morning. - Again, not checking ppms in or out anymore, she's ready Day 121- Day 73- 11/12 - Gave 1 liter of water Day 122- Day 74- 11/13 - Gave her last liter of water this morning - Tis her last day! Will cut this evening.

Day 22, 11th of October 2021: Helllooooo there. 1st day of the 4th week. Let's see how it goes.... we crazy... So: Mastodon Kush absolutely killing it.... just grow and grow another LST is performed. So one goes that way another one another way lol. Healthy and beautiful 😍 RQS girls both of them do well Cookies Gelato amazing lady.... LST performed. And no complain also about Northern Light neither.... She also deserved an LST training. And finally Spliff Seed girls: Afghani Gold and Spliffs Strawberry are one week younger but they pickin' up quick so soon it can not be seen. Anyways all good yes they grew a lot. CBD Medi Kush is amazing I fimmed a bit by accident of the top "head" no problem at all and again continued with LST. She is a beast. Blue Berry seems to be a bit "pale" let's say light but apart from that she is more than okay. LST is done on her too... Afghani Gold I just love her chunky indica comin' I can see the leaves lol. She received her first LST as well. Spliffs Strawberry is alright as well so I am happy. Actually, not much has changed since last week I just set the lamp back to 125W so approx 40% of the max and the water intake is more a bit so: Lamp is set to be on 125W approx 40% of the full power..... Humidity stays on approx 60% Ventilation switches on in every 2 hours for 5 min.... Watering like 0.08l per day.... Fertilization also with Bat Guano, Biobizz family - BioGrow, BioBloom, TopMax... so these ones 2x a week.... Epsom salt.... 1x a week.... Ratio can be found above. That's all basically. Starting to produce buds , they need a feeding we just got t Rain so I'm going to wait a couple days. Hard to tell when to water being planted in Earth.

After 2 days the roof is getting of and the plant gets her first 250 ml of Water. The plant grows very vigorously and fast. Unfortunate the last day the plant begun to bend her first 2 leaves up and i dont realy know why. I will try something out and see where it goes.

The temperatures, humidity, and watering volume(if measured) in grow conditions are all averaged for the week. The pH is soil pH. Any watering done by me is well water which is 7.6 pH and 50° F. Any listed nutrients are ml/gallon of soil. Day 1 we have lots of sunshine and temperatures in the mid 80's F. I watered today about 1.5-2 gallons per pot directly from the hose. Well water is 7.6 pH and about 50°F. #1 and #2 got topped today. #2 was topped in the evening at 6.30 p.m. I waited for it to get a little more space between the nodes, so there's a bit of stem above the node. This will help it to not split. Day 2 we had thunderstorms and rain in the morning followed by sunshine. The #3 plant was topped today. Day 3 we had lots of sunshine, temps in the mid 80's. That evening we had thunderstorms and rain which became severe that night. Hail was in the forecast but we missed it 🙏. I noticed the number 1 plant was growing crooked. I had moved the pots to get more sunlight and the one was uneven. I did some moving and adjusting today and hopefully it will straighten up. Day 4 we had rain in the morning followed by rain and thunderstorms intermittently into the night. Day 5 we had rains early morning and then cloudy most of the day. We have sunshine in the forecast for the weekend. 🙏🙏🙏 All pots were top dressed with 50ml of ferrous sulfate. Day 6 We had sunshine in the morning and afternoon. Clouds began to move in about 3.30p.m. Suddenly about 4:00 p.m. we had a severe thunderstorm that produced heavy rain and grape sized hail. Thankfully the hail only lasted a few minutes. There was some damage to this group, but only leaves. Day 7 we had sunshine, but cooler temperatures with highs reaching low to mid 70's. The plants began to show some Interveinal chlorosis in the new growth on day 5. I checked pH and it's above 7 in the pots. I added ferrous sulfate to counter this. On day 6 I noticed the #3 plant falling behind from receiving less direct sunlight. I've run out of space to accommodate the suns procession. I moved it to a better place for it to receive more sunlight. This week produced challenges. The topping was successful, but the position of the plants prevented some of them from receiving maximum direct sunlight. It took a few days to notice the affect on growth. We did not have much height gained this week. There was the issue with rising pH again, but this was promptly dealt with. We survived the challenging weather. Overall a successful week as lots of downward root growth was achieved. This will be evident next week as plants shoot up.

15 Day dry then trimmed using the Trim Bin. Curing with Grove bags, first harvest filled 3, 1/4lb grove bags. *Updating with clone pictures of this pheno in late flower labeled in diary as you look through, If I pop a new seed of this strain Ill start another diary

Que hay familia, vamos con la octava semana de floración de estas Candy Rain de Zamnesia, para el concurso POWER BUDS Plagron x Zamnesia CONTEST. Las flores van madurando progresivamente y desprenden unos aromas bastante dulces, también empiezan a tricomar bien y a madurar. Esta vez regué solo con agua controlando siempre el Ph, que ahora mismo lo dejamos en 6. Tragan alrededor de 0.8 litro por planta cada 48 horas.(3 riegos semanales). La temperatura máxima está en 22 grados y la humedad está entorno al 40%. Hasta aquí es todo, vamos viendo estas semanas como terminan de madurar nuestras flores.
