Yes, you'll be able to get a higher yield with the more plants you grow, but that is given that other factors are in place to. This can include the maintenance of the plants, feeding, genetics, and other factors that could ultimately affect yield. I'd say get a solid run-in with a couple of plants first before attempting to increase the total amount you are growing at a time.
That being said, given the dimensions of your tent, I would suggest if you are going for a large plant size, keep it to 1-2 plants. If you are going to be topping them and maintaining a smaller plant structure for their lives, then you can maybe squeeze 4-6 plants in a 40x40. Keep in mind you'll need to accommodate space for pots, humidifiers, lights, and whatever else you may need to throw in the tent.
So my advice, stick with one big plant for now, there's always the future where you can try more advanced and intensive runs than your first!
