My homework. Rubisco regeneration is intrinsically linked to nitrogen supply because Rubisco is a major sink for nitrogen in plants, typically accounting for 15% to over 25% of total leaf nitrogen. The regeneration phase itself consumes nitrogen through the synthesis of the Rubisco enzyme and associated proteins (like Rubisco activase), and overall nitrogen status heavily influences the efficiency of RuBP regeneration.Structural Component: Nitrogen is an essential building block for all proteins, and the sheer abundance of the Rubisco protein makes it the single largest storage of nitrogen in the leaf. Synthesis and Activity: Adequate nitrogen supply is crucial for the synthesis and maintenance of sufficient Rubisco enzyme and Rubisco activase (Rca), the regulatory protein responsible for maintaining Rubisco's active state. Nitrogen deficiency leads to a decrease in the content and activity of both Rubisco and Rca, which in turn limits the maximum carboxylation rate, Vmax, and the rate of RuBP regeneration Jmax, thus reducing overall photosynthetic capacity. Nitrogen Storage and Remobilization: Rubisco can act as a temporary nitrogen storage protein, which is degraded to remobilize nitrogen to other growing parts of the plant, especially under conditions of nitrogen deficiency or senescence. Nitrogen Use Efficiency (NUE): The allocation of nitrogen to Rubisco is a key determinant of a plant's photosynthetic nitrogen use efficiency (PNUE). In high-nitrogen conditions, plants may accumulate a surplus of Rubisco, which may not be fully activated, leading to a lower PNUE. Optimizing the amount and activity of Rubisco relative to nitrogen availability is a target for improving crop NUE. Photorespiration and Nitrogen Metabolism: Nitrogen metabolism is also linked to the photorespiration pathway (which competes with carboxylation at the Rubisco active site), particularly in the reassimilation of ammonia released during the process. To increase RuBisCO regeneration, which refers to the process of forming the CO2 acceptor molecule Ribulose-1,5-bisphosphate (RuBP) during photosynthesis, the primary methods involve optimizing the levels and activity of Rubisco activase (Rca) and enhancing the performance of other Calvin-Benson-Bassham (CBB) cycle enzymes. Biochemical and Environmental Approaches: Optimize Rubisco Activase (Rca) activity: Rca is a crucial chaperone protein that removes inhibitory sugar phosphates, such as CA1P (2-carboxy-D-arabinitol 1-phosphate), from the Rubisco active site, thus maintaining its catalytic competence. •Ensure optimal light conditions: Rca is light-activated via the chloroplast's redox status. Adequate light intensity ensures Rca can effectively maintain Rubisco in its active, carbamylated state. •Maintain optimal temperature: Rca is highly temperature-sensitive and can become unstable at moderately high temperatures (e.g., above 35°C/95F° in many C3 plants), which decreases its ability to activate Rubisco. Maintaining temperatures within the optimal range for a specific plant species is important. •Optimize Mg2+ concentration: Mg2+ is a key cofactor for both Rubisco carbamylation and Rca activity. In the light, Mg2+ concentration in the chloroplast stroma increases, promoting activation. •Manage ATP/ADP ratio: Rca activity depends on ATP hydrolysis and is inhibited by ADP. Conditions that maintain a high ATP/ADP ratio in the chloroplast stroma favor Rca activity. Enhance Calvin-Benson-Bassham (CBB) cycle enzyme activity: The overall rate of RuBP regeneration can be limited by other enzymes in the cycle. •Increase SBPase activity: Sedoheptulose-1,7-bisphosphatase (SBPase) is a key regulatory enzyme in the regeneration pathway, and increasing its activity can enhance RuBP regeneration and overall photosynthesis. •Optimize other enzymes: Overexpression of other CBB cycle enzymes such as fructose-1,6-bisphosphate aldolase (FBA) and triose phosphate isomerase (TPI) can also help to balance the metabolic flux and improve RuBP regeneration capacity. Magnesium ions, Mg2+, are specifically required for Rubisco activation because the cation plays a critical structural and chemical role in forming the active site: A specific lysine residue in the active site must be carbamylated by a CO2 molecule to activate the enzyme. The resulting negatively charged carbamyl group then facilitates the binding of the positively charged Mg2+ion. While other divalent metal ions like Mn2+ can bind to Rubisco, they alter the enzyme's substrate specificity and lead to dramatically lower activity or a higher rate of the non-productive oxygenation reaction compared to Mg2+, making them biologically unfavorable in the context of efficient carbon fixation. The concentration of Mg2+ in the chloroplast stroma naturally increases in the light due to ion potential balancing during ATP synthesis, providing a physiological mechanism to ensure the enzyme is activated when photosynthesis is possible. At the center of the porphyrin ring, nestled within its nitrogen atoms, is a Magnesium ion (Mg2+). This magnesium ion is crucial for the function of chlorophyll, and without it, the pigment cannot effectively capture and transfer light energy. Mg acts as a cofactor: Mg2+ binds to Rubisco after an activator CO2 molecule, forming a catalytically competent complex (Enzyme-CO2-Mg2+). High light + CO2) increases demand: Under high light (60 DLI is a very high intensity, potentially saturating) and high CO2, the plant's capacity for photosynthesis is high, and thus the demand for activated Rubisco and the necessary Mg2+ cofactor increases. Mg deficiency becomes limiting: If Mg2+ is deficient under these conditions, the higher levels of Rubisco and Rubisco activase produced cannot be fully activated, leading to lower photosynthetic rates and potential photo-oxidative damage. Optimal range: Studies show that adequate Mg2+ application can enhance Rubisco activation and stabilize net photosynthetic rates under stress conditions, but the required concentration is specific to the experimental setup. Monitoring is key: The most effective approach in a controlled environment is to monitor the plant's physiological responses e.g., leaf Mg2+ concentration, photosynthetic rate, Rubisco activation state, and adjust the nutrient solution/fertilizer to maintain adequate levels, rather than supplementing a fixed "extra" amount. In practice, this means ensuring that Mg2+ is not a limiting factor in the plant's standard nutrient solution when pushing the limits with high light and CO2. Applying Mg2+ through foliar spray is beneficial to Rubisco regeneration, particularly in alleviating the negative effects of magnesium (Mg) deficiency and high-temperature stress (HTS). While Mg can be leached from soil, within the plant it is considered a mobile nutrient, particularly in the phloem. Foliar-applied Mg is quickly absorbed by the leaves and can be translocate to other plant parts, including new growth and sink organs. Foliar application of: NATURES VERY OWN MgSO4 @ 15.0g L-1 in a spray bottle. Foliar sprays are often recommended as a rapid rescue measure for existing deficiencies or as a supplement during critical growth stages, when demand for Mg is high. Application in the early morning or late evening can improve absorption and prevent leaf burn. The starting point [of creativity] is curiosity: pondering why the default exists in the first place. We’re driven to question defaults when we experience vuja de, the opposite of déjà vu. Déjà vu occurs when we encounter something new, but it feels as if we’ve seen it before. Vuja de is the reverse—we face something familiar, but we see it with a fresh perspective that enables us to gain new insights into old problems.

Welcome Back!💚 Nach der zweiten Woche des Wachstums entwickelt sich eine stabile kleine Jungpflanze. Die THC-Victory entwickelt sich nun sehr rapide und es zeichnen sich bereits eine schöne längliche Blattform ab. Da es sich um eine reine Sativa Kreuzung handelt, werde ich die Pflanze vermutlich bereits ab Wachstumswochen 3 toppen und sie dann auch allmählich trainieren um ihr höhenwachstum im zaum zu halten. Diese Woche habe ich noch weitere Komponenten der BioTabsNL Reihe eingefügt. Ich habe das Selicium Flash laut Angaben als Top Dress Düngung eingearbeitet und die Tabs in die Erde gesteckt. Dann nochmal alles mit Bactrex angegossen. Die Umgebungsgegebenheiten sind OK: ————— 🌞 Temp: 21°C 🌚 Temp: 18°C bis 19°C 💨 RH: 54% VPD: 0,85 kPa ————— Viele Grüße 👋

Que pasa familia, vamos a añadir la tercera semana de vida de estas Apple Fritter de RoyalQueenSeeds, empezamos con la temperatura que la tenemos entre los 21/24 grados, la humedad está entorno al 60%, el ph está en 6.2 , el foco actualmente lo tengo encendido 18 horas y la potencia es al 50% de lo que da el foco. También añado la gama de nutrientes de Agrobeta, que no falla, sin duda de la mejor alimentación que hay para tus plantas. Trasplantamos a macetas definitivas, 7 litros en este caso, se ven algo amarillas las hojas más antiguas pero es porque pedían ya un trasplante en nada recuperan. Hi Gromies, well sometimes I can be my own worst enemy seems I defoliated ( actually I'll blame the apprentice Urban grow cat 😺) He defoliated a little to much, it's hard to get good help these days! Anyway she got a bit of light burn directly under light bulb area. A mix of to much defoliation & I also turned light up to super lumen setting & being lazy I hadn't hooked the extraction fan & ducting up to the light ( it's an air cooled light shade) pump air from outside tent through sealed light shade & exhaust it outside. I hooked that up & raised the light around 20 cm & all is good now, some burnt leaves. She is bouncing back & grown more leaves. It may have slowed Bud development a little! I changed light cycle to 12.5 hours of darkness 11.5 hrs on to try & kick her into gear , fool her into thinking the season is coming to an end with shorter daylight hours. I may increase dark period a little more bit by bit, experiment a little, I've read of other growers who give plants less light around week 6 till harvest. She is getting a faint lemony smell. Lollipoped some of the smaller secondary branches, any tips low down not receiving much light & don't look like they will get much bigger, other than that I'm leaving her alone, probably just try & seperate any close growing colas so we don't get any Bud rot. The strawberry gorilla is now chopped she finished a lot faster than the rest 9 weeks 6 days il add photos when she so dry iam guess 40-50g plants she was only small but super frosty beautiful smell.fur the otter the blue dream is getting close another couple weeks the others about 3 maybe 4 the west coast seems to be a very sensitive plants but never less she is stacking super frosty dense looking buds thays smell out of this world the og is also stacking

Me están pidiendo magnesio a tope, aparte de abonar con cal mag vía riego, también he abonado foliarmente... También he aprovechado y he añadido delta 9,... 5ml litro... Los tayos de las plantas y hojas algunas las tengo rojas, hay quien dice eso es la genética..., pero mis genéticas,no son rojas... Es falta de magnesio debido a la potencia del led... Les crea déficit de magnesio, también me pasaba con los LEDs chinos, pero la luz era rosa y no sé apreciaba ... Total que quiero indagar sobre el tema, yo creo que es carencia magnesio debido a la luz led ,la planta no da abasto,a asimilar tantos lúmenes y crea carencias... Mañana y pasado mañana abonaré foliarmente, solo con magnesio haber que pasa....

Día 37 (08/07) Ajustes de LST en todas las plantas! 🚀 Día 38 (09/07) Ajustes de LST en todas las plantas. Las ramas inferiores están creciendo como un cohete! 🚀😍 Riego con 250 ml H2O pH 6,5 Día 39 (10/07) Las plantas se muestran sedientas! Creo que el cepellón está lleno de raíces y piden el trasplante! El crecimiento no se ha visto afectado de momento Riego con 500 ml H20 pH 6,5 Día 40 (11/07) Paso a regar con 500 ml / día, ya que hace mucho calor (30 ºC) y efectivamente las plantas ya han copado la maceta de raíces OnionOG y KS1: Las ramas de nodo inferior se han quedado enanas tras el entrenamiento, de modo que las elimino y aplico canela en polvo en los cortes Riego con 500 ml H20 pH 6,5 Día 41 (12/07) Riego con 500 ml H20 pH 6,5 Clones! Mi experimento vuelve a demostrar que NO necesitas complejos productos de clonación ni sistemas para obtener clones! Hace 15 días puse 2 clones con el tallo sumergido en miel durante 5 minutos en una maceta de 400 ml con PRO-MIX HP BACILLUS + MYCORRHIZAE He mantenido la humedad alta con una cúpula de humedad casera hecha con una botella de agua PET de 5 litros cortada por la mitad y pulverizando a diario. Han estado los 15 días en mi terraza con luz indirecta, y temperaturas que rondan los 30 - 32ºC durante las horas centrales del día Hoy los he trasplantado a una maceta de 1 litro porque ya habían formado raíces y empezaban a formar nuevas hojas! 🐥🐥🚀🐥🐥 Día 42 (13/07) Riego con 500 ml H20 pH 6,5 Día 43 (14/07) Trasplante a maceta definitiva de 21 litros de ROOTPLUS Pot de GSKOREA GLOBAL! Estas macetas son una maravilla! Proceso de trasplante: Se prepara con 17,85 Litros (85%) de sustrato PRO-MIX HP BACILLUS+MYCORRHIZAE + 1,05 Litros de Humus de Lombriz (5%) + 2,01 Litros de Insect Frass (10%) + 210 gramos de Earth Vibes Super Soil (10 g/L substrato) Se llena la maceta de sustrato con las manos (limpias) y rompiendo los trozos más gruesos, para que el sustrato esté aireado y esponjoso, sin presionar Se coloca una maceta vacía de 6,5L para que quede la forma perfecta de la maceta donde están actualmente Se espolvorea la parte proporcional de la probeta de microorganismos sobre el agujero de trasplante Se saca la planta de su maceta actual (bonitas raíces 😍) y se coloca en la maceta final Se riega muy lentamente hasta percolación profunda con H2O EC 0,5 pH 6,5 Una vez asentada, complemento con un riego de 500 ml con 25 ml/L de Humus de Lombriz Liquido Se coloca mulch (acolchado) de paja para evitar traspiración excesiva y cuidar a los microorganismos del suelo A ver como reacciona al trasplante! Dernière semaine de croissance. Changement de solution fait le dimanche 22/11 car le ph commençait à descendre en dessous de 6.2 et cela faisait 1 semaine que j'avais fais le précèdent changement. Le PH lors du changement était de 6,8. Il est monté jusqu'à 7.5 mais je laisse le filtre faire son oeuvre. Il est actuellement à 7.1 et continue de descendre naturellement. carence en fer. Ajout d'une dose supplémentaire de Sensi CalMg Edit au 25 : Malheureusement ce que je sentais venir est arrivé, saturation en N. Le Ph et l'EC continuaient de baisser tous les deux. Les parties basses des plantes étant sombre et ayant une carence en fer sur la partie haute (puisque élément mobile), j'en ai déduit que c'était une carence en fer induite par une surfertilisation en N. J'étais à un EC de 1, je suis revenu à 0,8 le mardi 25 au soir. La bioponie est très capricieuse au niveau de l'azote. Le mercredi 26 le ph remonte ce qui signifie qu'on a stopé le processus de dégradation du N. Je devrais changer la solution mais je dois passer bientôt en flo donc j'ai enlevé de l'eau et rajouter, histoire de tenir jusqu'au prochain changement de solution qui est prévu samedi sauf si la situation dégénere de nouveau avec le PH. On Wednesday the 26th the ph goes up which means that we stopped the N degradation process. I should change the solution but I have to float soon so I removed some water and added more, to hold until the next change of solution which is planned for Saturday unless the situation degenerates again with the PH. Flowering day 74 since time switch to 12/12 h. Hey guys :-) Finally the time has come . The lady was harvested and hung upside down in the dry tent 😍. Before that, she stood in the drying tent in complete darkness for 2 days and the large leaves were removed. The small leaves stay attached during the drying process so that they protect the buds and allow them to dry more slowly and evenly :-) . In the dry tent, the temperature is 18-19.5 degrees and the humidity is 54-58% 👍. In this environment they are now allowed to hang for 10-14 days before they are fine-tuned and trimmed clean by hand. then after trimming they come to ferment for 6-12 weeks in dark jars with bobeda packs 62% to keep the perfect humidity. After a few weeks, the Boveda packs 58% are used so that they can slowly dry out :-) . Of course there is one last update after trimming. Until then, I wish you a lot of fun with the update. 2/9 - 4 gal RO 2/10 - 5 gal RO 2/11 - Feeding nutrients plus cal/mag, 5 gal 2/12 - Feeding nutrients, 5 gal 2/13 - FINALLY FIGURED OUT MY ISSUE! The ph within the soil was too low. RO only at 6.5ph 2/14 - RO only 6.8ph 2/15 - Feeding nutrients, 4 gal at 6.7ph Monday 2/15 was one of the coldest days we've had, our winter storm took our power that evening. I was ok with going without lights, the plants would have made it through a day or two. However I was not prepared for below freezing temperatures. Overnight my plants froze to death. I went without power for another 36 hours at below freezing temperatures. There was no way I could save them now.

Starting week 11 the heat wave is gone and the conditions lately have been perfect! The bud are forming nicely and the smel is strong but my diy carbon filter does a great job! Sticking with the same nutes and ph around 5.8. One of the two is slower than the other but comming along just fine
