After they finish stretching, which usually takes 3-4 weeks from when you flip the lights to 12/12, you'll want most of what's under the trellis to be lollipopped. This will make the plants push all their energy to the top canopy and make big ole buds.
Doing it this way keeps you from having to do much defoliation, since you won't have any nuggets trying to form underneath your main canopy. You'll only need to worry about leaves covering buds sticking straight out of the trellis, which doesn't happen too much.
Make sure you do remove leaves that are covering the majority of a bud site, though. There shouldn't be many, but there will be a few. Do this at the same time you lollipop, after the stretch that 12/12 triggers is finished.
